Defending champions Chiefs will host Komphela's former team at FNB Stadium, with kick-off at 19h30.
“We have a plan well worked out for the match. The message is clear, and I reminded the guys that this trophy is ours and we have to go out there and defend it,” Komphela told Amakhosi's website.
The ex-Bafana Bafana defender added: “We will monitor the players overnight [Tuesday]. It is not easy at the start of the season to ascertain the fitness of the players. But, those that will be included in the team, will do the job for us. We are ready and looking forward.”
The match is Komphela's first official assignment after replacing Stuart Baxter at Naturena in June.
Bidvest Wits and Ajax Cape Town advanced to the semi-finals of the event on Tuesday with respective victories over SuperSport United and Orlando Pirates.
Wits beat SuperSport 3-1, while Ajax edged the Buccaneers 1-0 at Orlando Stadium.
