Juventus, who needed two penalties to get out of jail at home to Atalanta, welcomed back Giorgio Chiellini, Alex Sandro, Miralem Pjanic and Gonzalo Higuain.
The tone was set by the defending champions five minutes gone when Danilo curled home Pjanic's cut-back on the edge of the six-yard area.
A second goal arrived inside 11 minutes after Higuain ran behind the defence to get onto Pjanic's ball for a composed execution.
Wojciech Szczesny frustrated the hosts when he rushed off his goal-line to thwart Domenico Berardi who charged in on goal 28 minutes in.
However, Sassuolo finally found the breakthrough a minute later when Filip Duricic combined with Francesco Caputo's inch-perfect delivery to beat Szczesny.
A change from the visitors at the break saw Daniele Rugani replacing Chiellini who struggled with a shoulder problem. Soon after the break, Berardi levelled matters for the home side after curling a free-kick over the wall and into the top right-corner.
Roberto De Zerbi's boys went in front for the first time three minutes later when Caputo moved around the defence to profit from Berardi's cross.
Cristiano Ronaldo had a golden chance to bring his side level 61 minutes in, but the Portuguese forward scuffed Paulo Dybala's ball straight at Andrea Consigli.
Juve still levelled matters inside minutes later as a defensive error allowed Sandro in at the near post to nod home Rodrigo Bentancur's delivery.
Juventus were nearly behind again, but Szcezesny palmed Hamed Traore's low shot away with the follow-up from Jeremie Boga cleared off the line by Sandro in the final minutes.
It ended all square though, as the Old Lady drop points for the third game running ahead of Monday's clash with Lazio.
