Man City were reported to be the most likely destination for the Argentine forward, and Mourinho has since claimed that the superstar could only sign a for a club that doesn't respect the Financial Fair Play Regulations (FFP).
Pep Guardiola's side were recently fined by UEFA for breaching FFP regulations but had their two-year European ban overturned, which the Spurs manager had criticised previously.
"(Would I sign Messi) for my (online football manager simulation game) Top Eleven team? No, not for Tottenham. Just for Top Eleven," Mourinho said to Joe.co.uk during a promotional video for Top Eleven.
"We respect Financial Fair Play at Tottenham. He can only go to a team that doesn't respect Financial Fair Play, so it's not Tottenham, for sure."
