Pirates lost 3-1 to the Egyptian giants on aggregate in the Final of the 2013 Champions League.
And Tinkler says his players are not lacking motivation ahead of the 18h00 kick-off in Soweto.
"We have done opposition analysis on them," Tinkler says.
"We showed the players a couple of games they played in the CAF Confederation and recently a game against Zamalek just to give them a general idea of how they play.
"The spirit is very high. I thought they will struggle a little bit after the draw against Polokwane City [in the League earlier this week] but that was not the case. Everybody knows the importance of the game and how big an occasion it is and everybody seems motivated.
"I am looking forward to it. This where you wanna be as a coach, you wanna be at this level. You wanna be playing CAF Champions League and Confederation Cup year in and year out. This is where you get challenged, tested, get noticed and I think that's an important thing and I am enjoying it.
"I don't have to motivate the players, they are highly motivated themselves because they know exactly who they are playing against. It's somebody they wanted to play for a long time. They are still disappointed with the results in 2013, and they have been talking to me that hopefully we meet Al-Ahly. They now have their chance."
Tinkler is hopeful lessons have been learnt from their continental heartache nearly two years ago.
"The lessons we learnt, here we ended updrawing the game after we gave away a silly free-kick outside the box and we got caught. Mohamed Aboutrika scored a great goal. We knew that was his strength and prior to the game we did state we shouldn't give away silly fouls and unfortunately that happened and that gave them an advantage.
"I don't think we want to find ourselves in the same position again going to Cairo knowing that you need that goal, leaving yourselves open because these teams are quality and they will punish you.
"It's vitally important that we try secure a win and most importantly we don't concede."
