The Brazilians went on to take the early lead when Lucas Costa Ribeiro showed some class by taking the ball in his own half before racing clear and slotting home from close range.
Sundowns' joy was short-lived as Dortmund equalised on 11 minutes when Ronwen Williams gifted the ball straight to Lukas Nmecha, who needed no second invitation to fire into an empty net.
The Bundesliga giants completed the turnaround on 34 minutes when Julian Brandt raced to the byline before picking out Serhou Guirassy, who rose well before heading into the back of the net.
Dortmund were not done there as Jobe Bellingham marked his first start for the club with a goal when he volleyed home from just inside the box after Williams cleared Brandt's cross.
Masandawana came out in the second half renewed and nearly got a second goal through a free-kick on the edge of the area by Teboho Mokoena, but it clipped onto the side-netting.
The Black and Yellow added to their tally ten minutes in when Khuliso Mudau directed a first-time cross from Daniel Svensson into his own net.
Sundowns halved the advantage in the 62nd minute as Iqraam Rayners pounced on a loose ball off the upright following a free-kick from Mokoena and bundled in his second goal of the competition.
Masandawana made a triple substitution nine minutes later and introduced Jayden Adams, Thapelo Morena and Arthur Sales, with Ribeiro, Mokoena and Matthews removed.
The South African champions continued to battle and grabbed a third in the last minute when substitute Lebo Mothiba side-footed in from close-range to open his account for the club.
There would be no dramatic comeback though, as the European giants held on in the closing stages to record their first win of the tournament.
