Spurs made two changes from their draw with Wolverhampton Wanderers, as Toby Alderweireld and Steven Bergwijn were recalled at the expense of Sergio Reguilon and Davinson Sanchez.
Leeds started the game with a lot of spark, creating good chances but were unable to convert them and Harry Kane nearly punished them in the 17th minute after latching onto a rebound on the blind side but his looping left-footed strike sailed slightly over the bar.
Leeds goalkeeper IIlan Meslier misplaced his pass and Bergwijn got a through ball and was brought down just inside the box in the 28th minute by Ezgjan Alioski, who gave the referee no other option but to point to the spot and the Video Assistant Referee (VAR) confirmed that the challenge was inches inside the box.
Kane stepped up to take the penalty and drilled a low shot through the middle, with Muslier going the wrong way to give Spurs a 1-0 lead.
Marcelo Bielsa's charges were leaving spaces behind and they paid the price for it with two minutes left of the first stanza after Son Heung-Min slotted a first-time strike at the near post from Kane's cross.
Leeds had a bad start in the second stanza as they went 3-0 behind six minutes inside the second stanza after Meslier could not keep out Alderweireld's header from a corner at the near post.
Bergwijn should have made it 4-0 for Jose Mourinho's troops just after the hour-mark after going clear on goal inside the box from a through ball but Meslier made a great save in a one-on-one situation.
The visitors piled on the pressure in the last quarter of an hour but Mourinho's men were happy to absorb the pressure and wait for their chances on counter-attacks.
Matt Doherty was sent off for Spurs in stoppage time for a second bookable offense, but the hosts comfortably clinched a 3-0 win that took them up to third on the table.
