The pair arrive at Upington on free transfers having parted ways with their respective teams.
26-year-old left winger Makgalwa became a free agent after being released by Sundowns ahead of the new season. While 27-year-old centre back Makume successfully negotiated a release from Maritzburg United following the club's relegation last season.
Meanwhile, Upington City are still looking for only their second win in the Championship after seven matches. They sit 13th on the log table and will have a chance to climb up when they take on Marumo Gallants on Saturday at Isak Steyl Stadium.
Kick-off is at 15:30.
