The teams will collide at Lucas Moripe Stadium on Friday, with kick-off set for 20h00.
Aces have been one of the surprise packages of the season as they hovered around the Absa Premiership summit before losing momentum in February.
"The team is hard at work," Booysen told his club's official website. "We know we are not going to play an easy opposition. Aces are doing well in the league and it is certainly going to be a tough one.
"However, we are cautious and looking forward to the big match. We appeal to our fans to come to the game and support us so that we win and advance to the next round."
Booysen, who returned to United on loan in January from Mamelodi Sundowns, also says a top-eight finish is a priority for Matsatsantsa.
"To be honest, we need to keep on working hard because there are few matches remaining before the season ends. Most of all we must fight to finish in the top eight and that will be good for our fans."
SuperSport are seventh in the Absa Premiership standings with 34 points from 25 games, just ahead of Chippa United on goal difference and two points in front of Ajax Cape Town.
