Following their 1-0 win over Free State Stars in their opening match, Matsatsantsa join Thanda Royal Zulu on six points and go into pole position thanks to their superior goal difference.
Goals from Elias Pelembe, Morgan Gould and Katlego Mashego ensured that the team from Tshwane went home with the three points in the bag, while Stars' Luis Renteria grabbed the home side's lone goal of the match from the penalty spot.
SuperSport got off to the perfect start, moving into the lead as early as the eighth minute when Mashego picked out Pelembe with a fine cross and the Mozambique international scored with a header.
Stars looked for an immediate reply when Godfrey Sapula shot at goal from a direct free-kick in the 10th minute, but United goalkeeper Dennis Onyango made a comfortable save.
Just prior to the half hour mark, Stars were gifted a way back into the game when the referee adjudged United's David Nkausu to have fouled Tsepo Ramokala in the 18-yard area and awarded a penalty.
Striker Renteria took on the responsibility and duly beat Onyango from the spot to bring his side level in the 29th minute.
However, the hosts' lead was short lived. Just two minutes later Gould rose highest to get on the end of Ricardo Katza's free-kick and headed Matsatsantsa back into the lead.
The defender's goal saw United leading 2-1 at half time, and just four minutes after the restart the visitors extended their advantage, as Daine Klate set up Mashego to score from 12 yards out and effectively assure the three points for Gavin Hunt's team.
The visitors soon went into cruise control and comfortably held Stars at bay for the remainder of the game, efficiently securing their second successive league win of the 2008/09 season.
