Mbappe, who has been linked with a big move to Real Madrid before the transfer deadline closes on Tuesday night, gave his side the lead with a header from a Di Maria cross in just the 16th minute.
Reims thought they had equalised through former Orlando Pirates player Marshall Munetsi in the 51st minute, but his goal was ruled out by the video assistant referee for offside.
Moroccan fullback Achraf Hakimi then provided an assist for Mbappe to double PSG's lead in the 63rd minute.
With 25 minutes left in the tie, Messi appeared on the sidelines in his new club's kit for the first time, and made his much-anticipated debut after signing for the Ligue 1 giants earlier this month.
With the forward replacing Neymar, the stadium made a resounding amount of noise and gave a standing ovation for the six-time Ballon d'Or winner as he jogged into position, according to BBC Sport.
Following the match, manager Pochettino spoke about Messi's display after his side kept up their 100% record in the 2021/22 Ligue 1 campaign with their 2-0 win.
"[Messi] is still far away from his best form but he has been training well and he will be fit in the next two weeks," he told Amazon.
"We are waiting for the best from him. I'm very happy that he made his debut. It was very important for him.
"He was good, from his first touches he gave the team peace of mind. It's good for him to start with a win.
"It was good to hear the crowd cheering for him, not just our supporters. It's something Leo has earned."
