The defender is regaining fitness after being sidelined by an ankle injury in January.
"He is back now," says Sundowns coach Pitso Mosimane, who has entrusted Thabo Nthethe with the armband in Mphahlele's absence.
"He has already given us 45 minutes and with the whole week I believe can give us 70 minutes, which is brilliant.
"If he can give us 70 minutes then we will go to Kinshasa with the advantage of having another extra player back into the team."
Mosimane, however, will be without suspended pair Bangaly Soumahoro and Khama Billiat for the trip to Kinshasa.
The Brazilians will next face Mbombela United in the Nedbank Cup last-16 on Saturday before visiting Vita inDemocratic Republic of Congo on April 9.
