Koena Mathopa should have put Santos ahead in the seventh minute when he missed a clear chance from inside the area.
Daine Klate sent a free-kick sailing over the Santos goal in the 12th minute, before Thando Mngomeni sent a long range effort into the arms of Supersport goalkeeper Denis Onyango.
Katlego Mphela should have opened the scoring for Supersport United in the 19th minute when he sent a header from close range straight at Santos keeper Lee Langeveldt.
The hosts dominated the rest of the half, but Santos defended well to ensure that the match remained goalless at the break.
The People’s Team gave a better showing in the second half and Isaacs should have handed them the lead in the 52nd minute, but the winger missed from six yards out.
However, the 20-year-old made no mistake three minutes later when he headed home a flick-on by striker Eleazar Rodgers from a Wayne Arendse long throw.
Supersport United substitute Katlego Mashego almost pulled his side level in the 59th minute, but the striker saw his powerful shot superbly saved by Langeveldt.
Daine Klate proved a prominent figure for the hosts as they showed more urgency in attack, unfortunately the winger’s teammates resorted to long range efforts, which never really troubled the Santos goal.
However, Tony Tsabedze almost handed Supersport a share of the spoils in the final minute of the game, but his spectacular long range shot was brilliantly saved by young Langeveldt.
