Mayambela hurt his shin in training, while Mashego strained a hamstring in the goalless draw with Golden Arrows last weekend and faces between one and two weeks on the sidelines.
Coach Teboho Moloi expects Mayambela to be back for their next clash with Platinum Stars at Nelson Mandela Bay Stadium on February 4.
"Key player for us and I am hoping to unleash him in front of the home fans when we welcome Platinum Stars. He has not featured this year but is a player that is very important to us for what he brings to the team," Moloi told the club's website.
'Tebza' also lamented the blow to veteran hitman Mashego.
"It is a huge blow to lose Katlego, I feel he was combining very well with Mxolisi Macuphu before we were forced to sub him early in the game. We are glad that he will not be sidelined for long."
Meanwhile, goalkeeper Brighton Mhlongo is nursing a toe complaint, but defender Ayabulela Konqobe resumed training this week after missing the Arrows game with a knee problem.
The Chilli Boys are eighth on the Premiership table with 24 points from 18 games - three points less than fifth-placed Maritzburg, who thumped high-flying Free State Stars 3-0 in Bethlehem last time out.
