Both South African sides play at home in the first leg on the weekend of July 11-13. The return legs will be played on the weekend of July 25-27.
The winners advance to the group phase, which starts in August.
CS Sfaxien defeated Orlando Pirates two years ago in the semifinal of the Champions League and last year beat Sundowns both home ad away in the Confederation Cup.
