Thomas Muenier gave the hosts the lead after putting the ball into his own net after seven minutes, before Juan Bernat restored parity four minutes later.
Bernat went from hero to villain when he was shown a straight red card for a foul on Nicolas Pepe after the 36th minute.
Lille then ran riot in the second half, with Pepe, Jonathan Bamba, Gabriel and Jose Fonte getting on the score-sheet.
